I wish to have a prototype panel of approx
100 * 250 mm. however this panel will contain 5 identical boards.

Should I send you the design once for you to panelize and rout? or should I make up a panel with the five boards?; in which case what details do you need for routing?

Just send us the single board. The work for setting up your panel is free.
If you would like to do it yourself, put a contour (small track) on any Layer.
